En los vinagres finales obtenidos el aminocido mayoritario sigue siendo la
prolina, pero ahora supone un 17% del total de aminocidos frente al 45% que
representaba en el sustrato de partida en las muestras F y un 57% frente al 62% inicial
en las muestras T. Segn lo descrito en la bibliografa (Valero et al., 2005; Palacios et
al., 2002; Erbe y Breckner, 1998; Llaguno y Polo, 1991b) en todos los vinagres de
vino la prolina es el aminocido mayoritario, pudindose considerar la presencia de ste
en el vinagre como un indicador de su procedencia vnica.
A pesar de los cambios experimentados durante la acetificacin el orden de
abundancia del resto de aminocidos se mantiene prcticamente igual al sustrato de
partida tanto en las muestras F como en las T.

4. CONCLUSIONES
1. Se ha adaptado y validado un mtodo para la determinacin de aminocidos en
vinagres de vino y vino en proceso de acetificacin mediante Cromatografa de
Lquidos empleando el agente 6-aminoquinolil-N-hidrosicuccinimidil carbamato
(AQC), obtenindose los siguientes resultados:
- En cuanto a la selectividad, la resolucin de los picos cromatogrficos
fue superior a 1.5 para la mayora de aminocidos.
- Se obtuvo una adecuada linealidad con unos coeficientes de correlacin
superiores a 0.992.
- El mtodo demostr tener una alta sensibilidad, con unos lmites de
deteccin entre 0.20 (Ala) - 45.73 (Arg) g/L y unos lmites de
cuantificacin entre 2.11 (Thr) - 70.05 (His) g/L, excepto para el caso
del triptfano, el cual tuvo unos lmites superiores (LDD: 110.37 g/L y
LDC: 280.75 g/L).
- Las recuperaciones fueron superiores al 80% para todos los aminocidos
excepto para serina y cistena.
- La precisin fue evaluada a dos niveles: repetitividad y precisin
intermedia. Estos niveles se determinaron tanto para el mtodo analtico
como para el proceso de derivatizacin, obtenindose en ambos casos
unos coeficientes de variacin acordes con los propuestos por el manual
AOAC.
2. Dada la alta sensibilidad del mtodo es necesario diluir las muestras. Despus de
valorar las concentraciones de aminocidos y amonio esperables en las muestras
tanto de vino como de vinagre y los intervalos de linealidad de dichos
compuestos, se opt por una dilucin del 25%.
3. Se han estudiado una serie de acetificaciones con cultivo superficial en madera,
y en ellos se observaron descensos estadsticamente significativos del nitrgeno
total.
4. En todas las muestras estudiadas, vinos y vinagres, el aminocido ms
abundante es la prolina seguido de la arginina; no detectndose, por el contrario
la asparagina, glutamina, cistena y triptfano. De todos los compuestos
estudiados el que las bacterias acticas utilizan en mayor extensin es la prolina.

5. Durante la acetificacin con cultivo superficial se observaron en la mayora de
los casos descensos estadsticamente significativos para: prolina, metionina,
asprtico, amonio, glicina, arginina, isoleucina, tirosina, y leucina. Los dos
primeros, prolina y metionina, mostraron descensos estadsticamente
significativos en todos las acetificaciones.
6. Ciertos aminocidos como la leucina, fenilalanina, valina y glicina presentan
evoluciones diferentes, ya que en las muestras F no presentan una tendencia
clara y en las T descienden mayoritariamente.
7. Los cambios en la concentracin de aminocidos y amonio a lo largo de la
acetificacin no suponen una alteracin en el orden de abundancia de los
mismos en los dos tipos de sustratos.
