Preparation of standard solution and dilution of standard solution
Objectives:

Prepare a standard solution of sodium chloride.

Dilute the prepared solution using the dilution law.

Material:
1.

Sodium chloride

2.

Beaker

3.

Glass rod

4.

Volumetric flask

5.

Glass funnel

6.

Distilled water

7.

Pipette

Procedure:
A. Preparation of a standard solution
1. You are required to prepare a standard solution of sodium chloride of volume 1000 ml and
concentration of 0.1 mol dm-3.
2. Calculate the mass of sodium chloride required to prepare the standard solution.
3. Weigh the calculated mass of sodium chloride in a small beaker.
4. Dissolve the sodium chloride in the beaker with small amount of distilled water. Stir the
solution with glass rod.
5. Transfer the solution into the 1000 cm3 volumetric flask with the aid of a glass funnel.
6. Add distill water to wash the beaker, the washing is transferred to the volumetric flask. Repeat
twice.
7. Carefully add distilled water until the graduation mark of the volumetric flask.

8. Stopper the volumetric flask and invert couple of times.

B. Dilution of standard solution


1. You are required to dilute the solution of sodium chloride prepared in A to produce a solution of
concentration 0.5 mol dm-3, 50 cm3.
2. Calculate the volume of sodium chloride solution from A and distilled water that is needed to
make the required dilution.
3. Pipette the required volume of sodium chloride solution from A into a conical flask.
4. Pipette the required volume of distilled water into the conical flask.
5. Mix the solutions thoroughly.

Iodine Mount Microscopic Examination on Feces Sample
Objectives:

Identify the morphology of cyst for Entamoeba histolytica and Balantidium coli

Identify the morphology of trophozoite for Entamoeba histolytica and Balantidium coli

Material:

Gloves
Tooth pick
Glass slide
Iodine
Disposal pipette
Cover slip
Microscope

Procedure:
1. Feces sample placed on biosafety cabinet sample
2. A small quantity of feces transfer on glass slide
3. A tooth pick use to flatten the sample and to make a smear on glass slide
4. Remove any fiber and particles
5. Smear added with a drop of iodine
6. Place a cover slip on top the smear
7. Observe under microscope from low power to high power
8. When cyst / trophozoite comes into view, identify under high power

9. When focus using high power, add few drops of immersion oil
10. Identify cyst / trophozoite based on special features and name the parasite

Objective

Identify the morphology of eggs for Paragonimus westermani and Ascaris Lumbricoides

Materials

Sputum sample
Gloves
Tooth pick
Glass slide
Normal saline
Disposal pipette
Cover slip
Microscope
:

1. Sputum sample placed on biosafety cabinet sample


2. A small quantity of sputum transfer on glass slide
3. A tooth pick use to flatten the sample and to make a smear on glass slide
4. Remove any fiber and particles
5. Smear added with a drop of saline
6. Place a cover slip on top the smear
7. Observe under microscope from low power to high power
8. When eggs comes into view, identify under high power
9. When focus using high power, add few drops of immersion oil
10. Identify eggs based on special features and name the parasite

Microscopic Examination on Urine Sample
Objective

Identify the morphology of eggs for Schistosoma hematobium

Materials

Urine sample

Gloves

Glass slide

Cover slip

Microscope

Immersion oil

Procedure
1. Urine sample placed on biosafety cabinet sample
2. Urine samples poured into test tube and centrifuge
3. After centrifuge, the sediment use to make a smear on glass slide
4. Place a cover slip on top the smear
5. Observe under microscope from low power to high power
6. When eggs comes into view, identify under high power
7. When focus using high power, add few drops of immersion oil
8. Identify eggs based on special features and name the parasite
