Routine maintenance procedures should be performed by the instrument user(s) in accordance with the schedule recommended by CEM Corporation as outlined below. Maintenance logs for twelve (12) months are provided in this manual for verication that the instrument is properly maintained for optimum operation and performance. Daily Maintenance 1. Inspect the instrument for any visible signs of leakage (liquid and/or stains from liquid) in the following areas: (Refer to illustrations on pages 2 and 3.) Spill Trays Under Ext1 and Ext2 Manifolds Under Rotary Valves Under Waste Fitting Manifold Under Main Valve Manifold (located toward bottom of instrument) Under Reaction Vessel Manifold Flat Surface Areas (especially under valves and ttings) Discover Spill Cup (especially around ttings and on bottom of spill cup) 2. Inspect the waste container to determine if it requires emptying. 3. Verify that proper nitrogen pressure is present (15 - 17 psi) (1.03 - 1.17 Bar) Weekly Maintenance 1. Replace the following lters: F1 and F2 (front of instrument) F3 (reaction vessel CEM P/N 172150)

Bi-Weekly Maintenance 1. Replace the following bottle lters: Dip Tube Filters on all Amino Acid Bottles (20) External Amino Acid Bottles (5) Filters in each Reagent Bottle (8) 2. Perform a system backush (page 7) on all amino acid and reagent bottle locations except DMF (Main Wash) and DCM (Secondary Wash).

Monthly Maintenance

Reagent Bottle

1. Perform a sensor calibration (page 10) on all liquid sensors. Export the new calibration values with a new le name to be saved on the computer hard disk. (Retain the previous calibration les.) 2. Verify reagent volume delivery. If necessary, perform a volume calibration (page 18). If calibration is performed, save the new calibration values in a new le on the computer hard disk. (Retain the previous calibration les.) 3. Inspect the reaction vessel for signs of particulate accumulation (particularly the inside top of the vessel). If necessary, clean the reaction vessel. Inspect all reaction vessel ttings. 4. Verify the performance of the ltered drain (page 24). Add 10 mL of DMF to the reaction vessel and perform a ltered drain to verify that the vessel empties within 10 seconds. 5. Review recent microwave method data to verify that when using 7 - 10 mL of reagent, the instrument can heat to 70 C within 75 seconds. Semi-Yearly Maintenance 1. Replace the reaction vessel (designed to use 100-200 mesh resins only) 2. Verify proper operation of the waste sensor. CAUTION In the event that the instrument is to be idle for a period of more than two (2) weeks, prepare the instrument as follows: Perform a system back ush. Remove all amino bottles and reagent bottle. Prior to use after the instrument is idle for more than two (2) weeks, Perform a system backush. Perform a sensor calibration. Perform a volume delivery calibration.

Backush WARNING Proper precautions must be taken during any routine maintenance procedure to avoid contact with reagents. Protective clothing, gloves and safety glasses should be worn as required. Refer to reagent manufacturers safety data sheets for specic guidelines for proper handling. 1. Remove all amino acid and reagent bottles. 2. Remove all amino acid and reagent dip tube lters. 3. Attach and tighten empty amino acid and reagent bottles (CEM P/N 167155) on all positions.

4. From the Main Menu, select Maintenance.

5. Select (Check) Select All. 6. Deselect (uncheck) Main Wash and Secondary Wash or DMF and DCM, based on the instrument software version, and any unused amino acid positions.

7. Select Backush.

Note: Ensure that steps 1 through 3 have been performed and that all amino acid and reagent bottles are tightened in position. 8. Select OK.

Note: The above message will appear for each amino acid and reagent as the tubing is ushed. Amino acids typically require approxmately one (1) minute to backush. Note: If necessary, select Stop to end the blackfush.

Note: As each amino acid and reagent tubing is backushed, it is deselected from the list and the Date Backushed is updated as illustrated below.

Once the backush process is completed, remove all amino acid and reagent backush bottles, and verify that each bottle contains approximately 3 - 5 mL of DMF.

10. Install new amino acid and reagent dip tube lters (CEM P/N 167485-M). 11. Install and tighten all amino acid and reagent bottles to be utilized for the next peptide. 12. Select Change Bottle for the following reagents to ensure that the tubing is properly primed: Main Wash (DMF), Secondary Wash (DCM), Deprotection, Capping, Activator and Activator B.

Protective clothing, gloves and safety glasses should be worn as required. Refer to reagent manufacturers safety data sheets for specic guidelines for proper handling.

1. From the Main Menu, select Maintenance.

2. From the Maintenance Screen, select Sensor Calibration.

3. Select (Check) Select All. 4. Deselect (Uncheck) LS2/3.

5. Select Calibrate.

6. Verify that a sufcient amount of DMF is available to ll the tubing. Select Yes.

6. Verify that 1) the nitrogen supply is turned on, 3) the DMF bottle contains a sufcient amount for calibration (approximately 500 mL), and 4) the reaction vessel is connected. 7. Select OK.

Note: The instrument will progress through several screens illustrated above as the DMF bottle is pressurized and the amino acid manifolds are lled. When these pre-calibration processes are complete, the sensor calibration begins. As each sensor is calibrated, an updated calibration value is displayed.

Note: Calibration will continue until all sensors are calibrated and new values are displayed (approximately 5 minutes). Upon completion, the above screen will appear.

Note: Upon completion of calibration, the following screen will appear.

Select OK.

Select Save. As each new calibration value is saved, it will be deleted from the above screen and the sensor will be deselected (unchecked). Once all values are saved, the following screen will be displayed.

10. Select Yes.

Note: The instrument will purge all tubing. Note: If necessary, press Stop to end the purging of instrument tubing.

11. Select OK.

12. Select Test Sensors. The System Diagnostics screen will be displayed.

Note: The Diagnostics screen will list the sensors to be tested and will provide a Sensor Test Started date. 13. Select OK. 14. If necessary, connect the DMF supply tubing and the waste bottle. Note: If, for some reason, the sensor testing must be terminated, select Cancel. 15. Select OK. The sensor testing requires approximately 5 minutes.

Note: As the sensor test(s) is successfully completed, a green check will be placed beside the applicable sensor and the instrument will indicate that the test passed. Once all sensors have been tested and passed, the following screen will be displayed indicating that the test is complete and the date and time of completion.

Note: If a sensor fails the sensor test, a red X will be placed beside the applicable sensor and the instrument will indicate that the test failed. It will also indicate the date and time of test completion and the specic sensor that failed. If a sensor test fails, contact the CEM Service Department.

16. Once the sensor test is complete, close the System Diagnostics screen and the software will automatically return to the Maintenance Sensor Calibration screen.

17. Select Export.

18. Using the computer keyboard, type the date of the sensor calibration. 19. Select Save to store the updated calibration data.

Volume Calibration WARNING Proper precautions must be taken during any routine maintenance procedure to avoid contact with reagents. Protective clothing, gloves and safety glasses should be worn as required. Refer to reagent manufacturers safety data sheets for specic guidelines for proper handling. 1. Remove the attenuator assembly from the cavity of the instrument. 2. Install the reaction vessel calibration stand (included in the instrument accessory kit) in the cavity of the instrument.

3. From the Main Menu, select Maintenance.

4. From the Maintenance Screen, select Volume Calibration.

5. From the Delivery selection menu, select the reagent volume to calibrate. The following reagents should be calibrated: DCM (Top), DCM (Bot), DMF (Top), DF (Bot), Deprotect, Capping, DMF Resin, and DCM Resin. (Refer to the screen on the next page.) CAUTION When using DCM, do not add Deprotect prior to draining the vessel or vise versa. Mixing these two chemicals causes solidication which can damage the instrument. Note: Only Boc customers should calibrate Add Cleavage Mix. After using Add Cleavage Mix, select Neutralization from the Maintenance Screen.

6. Select Calibrate.

7. Enter the amount of reagent to be used for calibration (10 mL recommended). 8. Select Add.

Wait for the instrument to deliver the selected reagent to the reaction vessel. Once the delivery process is complete, measure the volume in the reaction vessel.

10. Enter the amount of reagent delivered to the reagent vessel. 11. Select Calibrate.

12. Once the instrument completes the calibration of the selected reagent, the New Rate and New Flow values will be displayed. 13. Select Save. 14. Select Filtered to drain the calibrated reagent from the reaction vessel.

15. Repeat steps 3 through 12 for each reagent volume to be calibrated. CAUTION When using DCM, do not add Deprotect prior to draining the vessel or vise versa. Mixing these two chemicals causes solidication which can damage the instrument. 16. Once all reagent volumes have been calibrated, select Export to save the new calibration values.

17. Enter the date of calibration. 18. Select Save to store the reagent volume calibration values with the date of calibration.

Verication of Filtered Drain WARNING Proper precautions must be taken during any routine maintenance procedure to avoid contact with reagents. Protective clothing, gloves and safety glasses should be worn as required. Refer to reagent manufacturers safety data sheets for specic guidelines for proper handling. 1. Remove the attenuator assembly from the cavity of the instrument. 2. Install the reaction vessel calibration stand (included in the instrument accessory kit) in the cavity of the instrument.

4. Place 10 mL of DMF in the reaction vessel. 5. From the Maintenance Screen, select Main Wash or DMF (based on software version) from the second drop-down menu positioned under Reagent. 6. Select Add.

From the drain options on the Maintenance Screen, select Filtered.

As the DMF is draining from the reaction vessel, count the time (seconds) required to drain the 10 mL of liquid from the reaction vessel. CAUTION Drain time should not exceed 10 seconds. If the liquid fails to completely drain within 10 seconds, replace the reaction vessel (CEM p/n 167260).

Maintenance Records The following pages are provided for a monthly record of required maintenance procedures. As each procedure is completed weekly, monthly, semi-yearly, etc. as indicated on the appropriate record, either a check, date or the users initials should be entered on the record. The blue areas of each record require no action. In the event that a CEM Service Representative is required on-site, these records should be available for inspection to ensure that the required maintenance has been performed, ensuring maximum instrument performance.
